Elutriates obtained from the microcosms according to the proto-col EPA-823-B-01-002 [38] were used as analytical matrix. Briefly,40 mL distilled water were added to 10 g samples and the mix-ture was mechanically shaken for 1 h and centrifuged for 10 min at3500 rpm; the resulting supernatant was used as elutriate. Elutri-ate from solely RH was also analyzed for determining the intrinsictoxicity of this matrix. Ecotoxicological tests were performed intriplicates, using glass vials of 25 mL of capability and moderatelyhard reconstituted water without B12 vitamin complex for dilu-tions. A number of 5 daphnid neonates of less than 24 h were placedin each vial and then exposed to 10 mL of proper dilutions from theelutriates. The acute tests were performed in darkness at 23 ± 1◦C.Immobility at 24 h and 48 h was the bioassay endpoint, assumedas equivalent to mortality. EC50, the concentration producing 50%of immobilization in the daphnids, was determined using the TOX-CALC – Toxicity Data Analysis Software from Tidepool ScientificSoftware. In addition, toxicity units (TU) of the different sampleswere calculated according to the expression: TU = (EC50)−1× 100.3
