Estoy traduciendo un expediente farmacéutico de unos comprimidos / tabletas proveniente de la India. En la sección de pruebas para detectar la presencia de Salmonella, dice:

"Secondary test:

i) Subculture a suspected colony to Triple Sugar Iron Agar by first inoculating a surface of slant and **the deep inoculation in slant**. Incubate at 35°C to 37°C for 18 to 24 hrs.

ii) The formation of acid and gas in the stab culture with or without blackening of media and absence of acidity from the surface slant indicates the presence of Salmonella and vice versa.

iii) Precise confirmation may be carried out by appropriate biochemical and serological tests. The product passes the test if colonies of the type described do not appear or if the confirmatory biochemical and serological tests are negative.

iv) Carry out the control test with Salmonella organism prepare from 24 hr old culture. The test is not valid unless the results indicate presence of Salmonella in control test.

Limit: Absent."

Slant se traduce como agar inclinado. Se puede sembrar en forma superficial como una placa de Petri y/o en forma profunda, es decir "pinchando" el agar con el asa, o mejor una punta. Así se siembran inóculos en contacto con el aire, y sin contacto con el aire.
