Source text - English Purpose of Study
This study will help us learn more about a newly discovered vaccine drug, DSP-7888 Dosing Emulsion (or simply DSP-7888), that is being tested in people with your type of cancer. This drug may help your immune system to fight against cancer cells that have a protein called WT1. It is hoped that the vaccine will stimulate your immune system to kill or slow down the growth of cancer cells.
The main purpose of this study is to determine the highest dose of DSP-7888 that can be safely given to patients with your type of cancer. This study is not designed to test whether DSP-7888 is effective in treating your type of cancer.
DSP-7888 is an investigational drug, which means that it is still being studied and that research doctors are trying to find out more about this drug. It also means that the FDA (U.S. Food and Drug Administration) has not approved DSP-7888 for use in patients, including in people with your type of cancer, although it has allowed the drug to be tested in clinical studies like this one.
HOW MANY RESEARCH PARTICIPANTS WILL TAKE PART IN THIS STUDY?
The study Sponsor expects to enroll up to 60 participants nationally in this study.
WHAT IS INVOLVED IN THIS STUDY?
If you choose to participate in this research study, you will complete a “screening visit” that will consist of tests to determine whether you meet the study requirements. Some of the tests may have already been performed as part of the routine care you normally receive. Other required tests may be those that you would not otherwise receive, or would normally receive at a different time.
Your study doctor will discuss the results of the screening tests with you and you will find out if you are eligible to participate in this study.
During this study, you will be given injections of a new investigational drug, DSP-7888, under your skin. For the first month, you will receive injections of DSP-7888 once a week. For the next 6 weeks, you will receive injections of DSP-7888 once every 1-2 weeks. Thereafter, you will receive injections of DSP-7888 once every 2-4 weeks. The Sponsor of the study, Boston Biomedical, Inc., will provide the DSP-7888.
Your study treatment may stop if you have a side effect that isn’t acceptable or if your disease gets worse. You might also stop study treatment for other reasons, which your study doctor will talk with you about.
The dose of the DSP-7888 vaccine that you receive will depend on the number of other research participants that have been enrolled in this study before you and how well they have done after taking DSP-7888.
Depending on the dose to which you are assigned, you will receive between 2 and 10 injections on every dosing day (see below for dosing schedule).

Other Important Parts of the Study
Typing for HLA Class I and Class II
HLA (human leukocyte antigen) status is required for participation. HLA typing should be the first procedure performed. Human leukocyte antigen is a substance located on the surface of white blood cells. This substance plays an important role in the body’s immune response.
Biopsy of tissue for research participants with solid tumors
If you have a solid tumor, archival tissue (tissue obtained from a previous surgery or biopsy), is required. If archival tissue is not available, a baseline tumor biopsy at study entry is mandatory. This tissue would be collected for scientific research purposes; in order to evaluate the tissue in a laboratory for something called a “biomarker”. A biomarker is a substance, such as a chemical, that can be identified in tumor tissue through a laboratory test. Researchers hope to find biomarkers in order to help them learn whether there are any patients that would do particularly well with treatment with DSP-7888. Signing this consent means that you consent to a biopsy at the beginning of the study unless you have archival tissue available.
Mandatory bone marrow aspiration for research participants with AML or MDS
Bone marrow aspirates are mandatory for research participants with AML or MDS before you begin the study and at regular intervals during the study to measure your disease. If you have AML or MDS and consent to this study, you are consenting to these bone marrow aspirates.
Consent for Optional Biopsy
As an optional part of this study, the study Sponsor would like to obtain a tissue sample of your tumor after you have finished study treatment (that is, after you have received the last dose of vaccine). This tissue sample would be obtained by a biopsy of your tumor. This biopsy is completely optional. Information about this optional biopsy will be addressed later in a separate consent form. You can participate in the main study even if you do not consent to the optional biopsy part of the study.

Consent for Tissue Storage
After the study is over, and after the biomarker testing has been completed, it is possible that the samples and tissues collected during this study could be useful to other researchers. These samples could include blood, urine, biopsy samples, or archival tissue samples. You can give permission for these samples to be stored for research that might occur in the future, after this study is over. The exact nature of these potential research studies is not known: they could involve cancer or another disease entirely.
Consenting to “storage for possible future use” is optional. You can decline to store your tissue for possible future use and still participate in the study. More information about storing your blood and tumor tissue for potential future use will be addressed later in a separate consent form.
Please note – this is different from the work that will be done with your “archival tissue” described above. The archival tissue work is for biomarkers related to the study drug. By consenting to the study, you are allowing the biomarker work to be done if the archival tissue is available. If you decide to decline permission for future storage of your archival tissue, then the tissue will simply be returned after the biomarker work is done.

Source text - English This study will test the safety and immunogenicity of the gp120/NefTat/AS02A vaccine candidate in individuals with chronic HIV-1 infection successfully treated with HAART. The rationale for this study is based on previous scientific experiments, including data indicating that this vaccine can elicit strong HIV-1-specific T cell immune responses in humans and monkeys and lead to a retardation of HIV-1 disease progression in animal models of HIV-1 infection.
The HIV vaccine to be administered during this study consists of three recombinant HIV clade B viral antigens: the envelope glycoprotein gp120 and two regulatory proteins, Nef and Tat.The antigens are formulated in a proprietary adjuvant, AS02A, comprised of two immunostimulants in an oil-in-water emulsion (gp120/NefTat/AS02A). The vaccine and the adjuvant are manufactured and provided for the study by GlaxoSmithKline Biologicals, Rixensart, Belgium. The drugs will be given by intramuscular (IM) injection at a standard dose of 20 mg together with 0.5 ml of the AS02A adjuvant.
Twenty HIV-1 infected individuals will be randomly enrolled into three different study groups, receiving either the gp120/NefTat/AS02A vaccine (10 individuals), the AS02A adjuvant alone (5 individuals) or a placebo (5 individuals). After obtaining informed consent, subjects will have a history and physical exam performed and have laboratory tests to confirm they meet all inclusion and exclusion entry criteria. Women of childbearing potential will have a pregnancy test prior to each injection of the investigational product. Injections with vaccine, adjuvant alone, or placebo will then be performed at weeks 0, 4, and 12. Study participants will undergo close monitoring after each vaccination. Blood samples will be obtained for immunological assays at study baseline (2 times) and weeks 2, 4, 6, 12, 14, 24, and 48. All patients will maintain their antiretroviral treatment regimen during the entire study period.
Primary Outcomes: Occurrence, intensity and relationship of any local and general signs and symptoms during a 7-day follow-up period (i.e. day of vaccination and 6 subsequent days) after each vaccination; Changes in the frequency (%) of IL-2 secreting CD4+ T cells, in response to at least one vaccinal antigen, at two weeks post the third vaccination.
