the "no." isn't necessary and is usually omitted; the comma is unnecessary and a matter of style - I usually omit it for brevity

sorry, in a document listing work to be carried out on properties, the "no." would be included - but in writing an address it's not nec.

I don't think in English it's necessary to put nº. I think a very common way to write it in English is simply 4xx yyy Street. However, if rua is really part of the street name and you don't want to translate it, then 4xx rua ..., as you have it. I would use the comma if I changed the order of it and said rua xx, no. 4xx, Arranged in that order I think you need the no. and comma.
