7.3.2 Gewinnung von Plasma
a. Die Monovetten mit dem EDTA-Blut werden 10 Minuten bei 1500 g bei Raumtemperatur zentrifugiert. Bei der Multifuge 1 S-R mit ***Bremsauslauf*** auf Stufe 1, bei der Megafuge 1,0 R ab 50 g mit Bremsabschaltung.
b. Der Plasmaüberstand wird vorsichtig, mit Abstand zu den Zellen, in 15 ml Falkonröhrchen überführt und 10 Minuten bei 3000 g bei Raumtemperatur mit Bremse zentrifugiert.
c. Es wird 1 ml Plasma pro Aliquot unter Beachtung des Abstands zu dem Zellpellet abgenommen und in Cryotubes pipettiert. Dabei wird das gewonnene Plasma auf Abweichungen vom Standard und Besonderheiten (Siehe 7.5) untersucht.
d. Die Plasma-Aliquots werden bei -20°C in einem Gefrierschrank eingefroren. Am Ende des Arbeitstages werden die Aliquots im Gefrierschrank bei -80°C zur endgültigen Lagerung eingefroren. Die Umlagerung erfolgt in einer Styroporkiste mit Trockeneis.

To gently stop a centrifuge, particularly for use in blood transfusion procedures including plasmapheresis, a centrifuge motor is first braked by reverse current application with high braking torque, to reduce the speed thereof from operating speed to less than 1000 rpm, for example between 800 to 500 rpm; and then braked under dynamic braking conditions, preferably under continuously variable torque conditions, until it is about stopped, for example at close to 50 rpm, and then permit it to coast to a stop.
